We report on the first multiplex preparative separation by mass spectrometry of bio-organic molecules in the 200-350 Da mass range that is typical for synthetic drugs. A five-component mixture consisting of two di- and three tripeptides has been separated by mass using a specially designed mass spectrometer. The instrument for preparative separations consists of an electrospray ionization (ESI) source, ion transfer optics, an electrostatic sector, and an inhomogeneous-field magnetic mass analyzer that achieves linear mass dispersion of ion beams. Protonated peptides produced by electrospray were separated, nondestructively landed on a 16-channel array of dry collector plates, and reconstituted in solution. The preparation procedures and the instrumental conditions have been optimized to maximize the ion currents. The significant features of the special mass spectrometer are high ion currents and simultaneous separation and collection of mixture components. 相似文献

Two types of thiophene-capped [2]rotaxanes, i.e., bithienyl (2T)- and bis(3,4-ethylenedioxythiophene)-yl (BEDOT)-capped [2]rotaxanes, were synthesized. The electron-deficient cyclophane of cyclobis(paraquat-p-phenylene) (CBPQT4+) was used as a macrocycle. Association constants for inclusion complexation of 2T- and BEDOT-derivatives with CBPQT4+ were obtained by 1H NMR titration. Due to the donor-acceptor charge transfer absorption band, 2T- and BEDOT-capped [2]rotaxanes have red and green colors, respectively. On the basis of electrochemical analysis, we confirmed that only BEDOT-capped [2]rotaxane is a promising candidate for [3]rotaxane synthesis through oxidation coupling of the thiophene unit. 相似文献

用石英弹簧法和示差扫描量热法 (DSC)、红外分光光度计 (FTIR)研究了苯、乙醇、丙酮、醋酸乙酯和1,2 二氯乙烷五种溶剂在端羟基聚丁二烯 丙烯腈共聚物为软段的聚氨酯中的溶解和扩散行为 .结果表明所有溶剂在丁腈聚氨酯中的扩散均为非费克扩散 ,且随着溶剂蒸汽压增大偏离费克扩散的程度增大 .相同相对蒸汽压下 1,2 二氯乙烷和醋酸乙酯偏离费克 (Fickian)扩散的程度较大 ,而乙醇、丙酮和苯则较小 ,这主要与它们和丁腈软段溶解度参数的极性分量和氢键分量有关 .1,2 二氯乙烷和苯在HTBN PU中的溶解度较高 ,而乙醇 ,醋酸乙酯和丙酮较低 ,主要与它们和丁腈软段溶解度参数的色散分量有关 .所有溶剂均表现出近似Flory Huggins型等温吸收曲线 .红外表明吸收溶剂后 ,氨基甲酸酯基团的氢键化程度有不同程度的下降 ,和溶剂与之形成氢键的能力大小有关 .力学性能表明非极性溶剂苯对材料的力学性能影响较小 ,而乙醇 ,醋酸乙酯和丙酮由于可与氨酯基团形成氢键 ,对原HTBN PU中氨酯键氢键的破坏大 ,力学性能下降大 相似文献

Silica gels retaining double pore structure in the size ranges of micrometer and nanometer have been applied to the rod-shaped
monolithic column for liquid chromatography. The macropore structure was designed by controlling the phase separation process
induced by the hydrolysis and polycondensation of alkoxysilane, whereas the mesopore structure was tailored by the solvent
exchange treatments on wet gels. The size exclusion chromatograms on polystyrene standards exhibited almost similar features
for octadecyl-modified rod and conventional packed beads columns. The dependence of plate height on the velocity of mobile
phase determined for amylbenzene was by far weaker in the rod column than in the packed beads column, suggesting that additional

A novel chiral selector, clindamycin succinate, was synthesized and first used as a chiral selector in capillary electrophoresis (CE). The chiral resolution ability of this kind of clindamycin derivation was studied by CE using some racemic drugs as model analytes. From the experimental results, it was found that both resolution and selectivity of the selector were dependent on the following parameters: concentration of chiral selectors, pH of the running buffer, temperature of the capillary column, applied voltage and organic modifier used. The results show that the chiral selector possesses high resolution toward some racemic drugs, including ofloxacin, chlorphenamine, tryptophan, propranolol, sotalol and metoprolol. Excellent chiral resolution of these tested drugs was achieved under the optimal conditions of 50 mM clindamycin succinate, 10% MeOH v/v, 50 mM Tris buffer, pH 4.0, at 22 kV and 20 °C within 25 min. 相似文献

The chiral separation of simendan enantiomers using capillary electrophoresis was studied with beta-cyclodextrin (beta-CD) as chiral selector. The influences of the concentration and pH of borate buffer solution, beta-CD concentration and methanol content in the background electrolyte were investigated. These factors were compared with those in an HPLC with beta-CD as chiral mobile phase additive (CMPA-HPLC). The quantification properties of the developed CE method were examined. A baseline separation of simendan enantiomers was achieved in the background electrolyte of 20 mmol/L borate buffer (pH 11.0) containing 12 mmol/L beta-CD-methanol (50:50 in volume ratio). The CE method is comparable with CMPA-HPLC in chiral resolution, although the optimal pH in CE (11.0) is much higher than that (6.0) in CMPA-HPLC. A convenient method is presented for the first time for the direct separation of enantiomers of a tris(2-phenylpyridine) iridium (III) and an analog substituted with long alkoxy chains on polysaccharide derivative-based chiral stationary phases by HPLC. Tris(2-phenylpyridine) iridium (III) was separated on the immobilized amylose 3,5-dimethylphenylcarbamate (Chiralpak IA) using hexane/CHCl3/CH2Cl2 (75:20:5) as an eluent, and the analog could be separated on the coated cellulose 3,5-dimethyl-phenylcarbamate (Chiralcel OD) and cellulose 4-methylbenzoate (Chiralcel OJ) using hexane/2-propanol (96:4) as the eluent. A flow solid phase extraction procedure based on biosorption of Pt(IV) and Pd(II) on Aspergillus sp. immobilized on cellulose resin Cellex-T was proposed for the separation and preconcentration of Pt and Pd before their determination by electrothermal atomic absorption spectrometry (ETAAS). The analytical conditions including sample pH, eluent type, flow rates of sample and eluent solutions were examined. The analytes were selectively retained on the biosorbent in acidic medium (pH 1) and subsequently eluted from the column with 1 mL of thiourea solution (0.25 mol L− 1 thiourea in 0.3 mol L− 1 HCl). The reproducibility of the procedure was below 5%. The limit of detection of the method was 0.020 ng mL− 1 for Pt and 0.012 ng mL− 1 for Pd. The method validation was performed by analysis of certified reference materials BCR-723 (tunnel dust) and SARM-76 (platinum ore). The developed separation procedure was applied to the determination of Pt and Pd in road dust samples by ETAAS.The applied biosorbent is characterized by high sorption capacity: 0.47 mg g− 1 for Pt and 1.24 mg g− 1 for Pd. 相似文献
